How do you know when to use the different kun and on sounds for this kanji? Can you say whichever one you want?

When referring to the color white as a noun, it is always しろ. If you are describing something as being white (i.e. using it as an adjective), it's しろい. In my experience, the on-yomi はく is pretty much only used in kanji compounds (e.g. 白銀), but be careful because it's not always read that way (see 白髪).
